FOXC1/2 Transcription Factor Activity Assay

The FOXC1/2 Transcription Factor Activity Assay from Assay Genie is a DNA-binding ELISA kit that quantifies the DNA-binding activity of forkhead box transcription factors FOXC1 and FOXC2 directly in nuclear extracts prepared from human, mouse, and rat samples.

FOXC1 and FOXC2 belong to the forkhead/winged-helix family of transcription factors with established roles in embryonic development, ocular morphogenesis, and lymphatic vessel formation. Mutations in FOXC1 are linked to Axenfeld-Rieger syndrome and primary congenital glaucoma, while FOXC2 alterations are associated with lymphedema-distichiasis syndrome. By measuring functional DNA-binding activity rather than total protein abundance, the kit supports research in developmental biology, ophthalmology, vascular biology, and oncology.

The microplate-based colorimetric assay uses a consensus forkhead response element immobilized on the plate to capture active FOXC1/FOXC2 from nuclear and cell lysates, followed by a specific antibody and an HRP substrate to generate an absorbance readout.
