FOXB1/2 Colorimetric Cell-Based ELISA

This cell-based ELISA kit from Assay Genie is designed to measure FOXB1/2 protein expression profiles in cultured cells. Using a colorimetric detection method at 450 nm with a dynamic range of > 5000 Cells, the kit is reactive with Human and Mouse samples and is intended for high-throughput applications in a 96-Well Microplate format.

The assay employs an indirect ELISA format in which FOXB1/2 is captured by FOXB1/2-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. The kit is lysate-free and can be used to measure relative FOXB1/2 levels in cultured adherent cells, as well as to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on FOXB1/2. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density for additional normalization.

FOXB1 is a DNA-binding transcription factor of the forkhead box protein family, localised to the nucleus, with roles in transcription regulation, somitogenesis, and neural development. The kit is therefore relevant to research areas including cancer, neuroscience, and stem cell biology, falling within the Epigenetics and Nuclear Signaling research field.
