Description

From Assay Genie, this Cell-Based ELISA Kit provides a sensitive, high-throughput, lysate-free method for detecting FosB protein expression directly in cultured cells. Detection is colorimetric, read at 450 nm, and the assay is reactive with human and mouse samples with a quoted dynamic range of greater than 5000 cells. The kit is supplied in a 96-well microplate format and is intended for storage at 4°C for up to six months.

The workflow uses an indirect ELISA approach: FosB is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. Because the readout is qualitative, two normalisation strategies are included. A monoclonal anti-GAPDH antibody serves as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density so that absorbance can be adjusted for plating differences. The kit is suitable for measuring relative FosB levels in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on FosB expression.

FosB is a transcription factor of the bZIP family that dimerises with Jun proteins to form the AP-1 transcription factor complex, and has been implicated in the regulation of gene expression related to chronic stress, addiction, and neural plasticity. The associated human protein (UniProt P53539; NCBI Gene ID 2354) has a reported molecular weight of 30,978 Da. Assay Genie positions this kit as a resource for research into addiction, psychiatric disorders, and neuroplasticity.
