FLI1 Colorimetric Cell-Based ELISA Kit

From Assay Genie, this cell-based ELISA kit provides a qualitative colorimetric readout for measuring FLI1 protein expression directly in cultured cells. The assay is read at 450 nm and is reactive with human and mouse samples, with adherent cells supported as a sample type.

Designed as a lysate-free, high-throughput workflow, the kit uses an indirect ELISA format: FLI1 is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies then catalyse a colour change upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, while a Crystal Violet whole-cell staining step allows cell density to be determined so that absorbance values can be adjusted for plating differences. The approach supports screening of the effects that various treatments, inhibitors or activators have on relative FLI1 levels.

The dynamic range of the assay is > 5000 cells. Storage is recommended at 4°C for 6 months. The product is placed within the Epigenetics and Nuclear Signaling research area and targets FLI1, a sequence-specific transcriptional activator whose dysregulation has been linked to conditions including certain leukaemias and autoimmune disorders, as well as to Ewing sarcoma via characteristic chromosomal translocations such as t(11;22)(q24;q12) involving EWSR1.
