Description

Designed by Assay Genie for the accurate and reliable detection of FHIT (Fragile Histidine Triad) protein levels, this Colorimetric Cell-Based ELISA Kit works with cell lysates and tissue samples from human sources. The assay is intended for use with adherent cells and uses a colorimetric detection method read at 450 nm.

The kit is a lysate-free, high-throughput, sensitive assay for measuring relative FHIT protein expression in cultured cells, suitable for screening the effects of treatments, inhibitors, or activators. It uses an indirect ELISA format in which FHIT is captured by FHIT-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method is provided for determining cell density. The dynamic range is > 5000 Cells and the assay is supplied in a 96-Well Microplate format with a storage stability of 4°C for 6 Months.

FHIT is a member of the histidine triad gene family, encoding a diadenosine 5',5'''-P1,P3-triphosphate hydrolase involved in purine metabolism, and is considered a possible tumor suppressor in specific tissues. Aberrant transcripts from this gene have been found in about half of all esophageal, stomach, and colon carcinomas. The kit is listed under the Cell Death research area and is intended for studies of cancer-related molecular mechanisms and therapeutic target identification.
