From QIAGEN, this kit enables high-speed preparation of first-strand cDNA directly from cultured cells without the need for RNA purification, with the entire procedure completed in just 45 minutes. It is designed for use in real-time, two-step RT-PCR and gene expression analysis directly from cells, making it well suited to experiments requiring rapid, high-throughput gene expression analysis.

The workflow follows a streamlined four-step procedure: cell wash to remove extracellular contaminants, cell lysis with integrated RNA stabilization, genomic DNA elimination using the included gDNA Wipeout Buffer, and reverse transcription. The synthesized cDNA is ready to use in real-time two-step RT-PCR, and the approach removes the need to design RNA-specific primers or probes. The kit integrates with optimized QuantiTect, QuantiFast, or Rotor-Gene kits for downstream real-time RT-PCR analysis, allowing several cell samples to be processed and analysed within a few hours.

Supplied reagents include Buffer FCW for effective removal of extracellular contaminants, Buffer FCP for efficient cell lysis and RNA stabilization, gDNA Wipeout Buffer (7x) for elimination of genomic DNA contamination, Quantiscript Reverse Transcriptase (an optimized blend of Omniscript and Sensiscript reverse transcriptases with RNase inhibitor), Quantiscript RT Buffer (5x) containing dNTPs and Mg2+, RT Primer Mix (a blend of oligo-dT and random primers), and RNase-Free PCR-grade water.
