Description

Assay Genie offers a Cell-Based ELISA kit designed to study the phosphorylation status of FANCD2 at serine 222 in cell-based assays. The kit provides a lysate-free, high-throughput workflow for measuring relative levels of phosphorylated FANCD2 in cultured cells, and can be used to screen the effects of treatments, inhibitors, or activators on FANCD2 phosphorylation.

The assay uses an indirect ELISA format: a phospho-specific primary antibody captures FANCD2 (Phospho-Ser222), and an HRP-conjugated secondary antibody catalyses a colorimetric reaction upon substrate addition, read at 450 nm. A monoclonal anti-GAPDH antibody is included as an internal positive control, and Crystal Violet whole-cell staining may be used to normalise absorbance values to cell density. The kit is reactive with Human, Mouse, and Rat samples, is suited to adherent cell work, and has a stated dynamic range of > 5000 cells.

FANCD2 is a DNA repair protein required for maintenance of chromosomal stability, accurate pairing of homologs during meiosis, and repair of DNA double-strand breaks by homologous recombination and single-strand annealing. It may participate in S phase and G2 phase checkpoint activation upon DNA damage. The phospho-Ser222 readout makes this kit relevant to researchers investigating DNA damage response pathways, genomic stability, and potential therapeutic targets in cancer and related genetic disorders.
