Description

From Assay Genie, this colorimetric cell-based ELISA kit is designed for the quantification of phosphorylated FAK (pTyr925) levels in cell lysates. The lysate-free, high-throughput and sensitive assay can detect FAK protein phosphorylation and expression profile in cells, and is suited to measuring the relative amounts of phosphorylated FAK in cultured cells as well as screening for the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on FAK phosphorylation.

Qualitative determination of FAK (Phospho-Tyr925) concentration is achieved by an indirect ELISA format: phospho-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition at 450 nm. The kit is reactive with human, mouse and rat samples, with a dynamic range of > 5000 cells, and is intended for use with adherent cells.

Phosphorylated FAK at tyrosine 925 is a key signalling molecule involved in cell adhesion, migration and survival, and its dysregulation has been implicated in various diseases including cancer and inflammatory disorders. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, while the Crystal Violet whole-cell staining method may be used to determine cell density. FAK is a tyrosine kinase of the FAK family required for cell migration and contact-dependent survival signalling, and increased expression has been reported in breast and other cancers.
