Description

From Assay Genie, this cell-based ELISA kit is designed to detect Focal Adhesion Kinase (FAK) phosphorylated at the Tyr861 residue. Suitable for adherent cells, the kit supports research into FAK signaling pathways involved in cell adhesion, migration, and proliferation, with relevance to disease contexts including cancer, cardiovascular disorders, and fibrosis.

The assay has reactivity for Human, Mouse, and Rat samples, a dynamic range of > 5000 Cells, and a colorimetric readout measured at 450 nm. It uses an indirect ELISA format in which target-specific primary antibodies are bound by HRP-conjugated secondary antibodies, allowing qualitative determination of phosphorylated FAK in cultured cells. The kit can be applied to measure the relative amounts of phosphorylated FAK and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on FAK phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining method may be used to assess cell density following the colorimetric measurement.

The target gene is PTK2 (Focal Adhesion Kinase 1), with chromosomal location 8q24.3, UniProt ID Q05397, NCBI Gene ID 5747, OMIM 600758, and Unigene Hs.395482. FAK is a non-receptor tyrosine kinase concentrated at focal adhesions, activated downstream of integrin clustering and Src signaling, and implicated in cytoskeletal organization, cell motility, and survival pathways.
