Description

A Cell-Based ELISA Kit from Assay Genie designed to detect phosphorylation of Focal Adhesion Kinase (FAK) at the tyrosine 576 site. The kit is a convenient, lysate-free, high throughput and sensitive assay that can detect FAK protein phosphorylation and expression profile in cells, enabling measurement of the relative amounts of phosphorylated FAK in cultured cells as well as screening for the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on FAK phosphorylation.

Qualitative determination of FAK (Phospho-Tyr576) concentration is achieved by an indirect ELISA format. FAK (Phospho-Tyr576) is captured by target-specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The HRP enzyme conjugated to the secondary antibody can then catalyze a colorimetric reaction upon substrate addition, measured colorimetrically at 450 nm. The assay is reactive with Human, Mouse and Rat samples and is suitable for Adherent Cells, with a dynamic range of > 5000 Cells. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts.

The target gene symbol is PTK2 and this is a Phospho-specific sub-type ELISA kit, sharing the broader FAK family biology of being a tyrosine kinase involved in cell migration and contact-dependent survival signaling, localizing to focal adhesions.
