Description

This cell-based ELISA kit from Assay Genie is designed for the accurate and sensitive detection of phosphorylated Focal Adhesion Kinase (FAK) at the Tyr397 site in cell samples, with high specificity and reproducibility for research into cell signaling pathways and cancer biology. FAK is described as a key regulator of cell adhesion and migration, and phosphorylation of FAK at Tyr397 is reported to activate downstream signaling pathways involved in cell growth and survival.

The kit is configured as a Cell-Based ELISA targeting FAK (Phospho-Tyr397) and is reactive with human, mouse, and rat samples. The recommended sample type is adherent cells. Detection is colorimetric and measured at 450 nm, with a dynamic range of greater than 5000 cells. The assay is lysate-free and high-throughput, using an indirect ELISA format in which FAK (Phospho-Tyr397) is captured by phospho-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the conjugated HRP to catalyze a colorimetric reaction upon substrate addition.

For data normalization, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and a Crystal Violet whole-cell staining method can be used to determine cell density so that absorbance values can be normalized to cell amounts. The target is encoded by the PTK2 gene (Gene ID: 5747, UniProt ID: Q05397), a cytoplasmic protein tyrosine kinase concentrated in the focal adhesions that form between cells growing in the presence of extracellular matrix constituents.
