Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the measurement of phosphorylated FAK (phospho-Ser843) levels in cell lysates. The assay uses a colorimetric detection method at 450 nm in a 96-well microplate format and shows reactivity with human, mouse, and rat samples. It is suited to adherent cell samples and has a dynamic range of greater than 5000 cells.

The kit operates as a lysate-free, high-throughput and sensitive assay that can detect FAK protein phosphorylation and expression profile in cells. It works via an indirect ELISA format in which phosphorylated FAK (phospho-Ser843) is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalising target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density for further normalisation.

Phosphorylated FAK (phospho-Ser843) is described as a key regulator of cell adhesion, migration, and survival, playing roles in cell growth, differentiation, and invasion. Aberrant FAK phosphorylation has been linked to cancer progression and metastasis, making it a relevant target for cellular signalling and pathway studies, cancer biology, and drug development. The target is encoded by the PTK2 gene (Gene ID: 5747; UniProt ID: Q05397) and belongs to the focal adhesion kinase family.
