Description

From Assay Genie, this Cell-Based ELISA kit is designed for the accurate detection of FADD phosphorylation at Ser191 in cell lysates, using a sensitive indirect ELISA format in which FADD (Phospho-Ser191) is captured by a phospho-specific primary antibody and detected via HRP-conjugated secondary antibodies with a colorimetric reaction read at 450 nm.

The kit is reactive with mouse samples and has a dynamic range of > 5000 Cells. A monoclonal GAPDH antibody is included as an internal positive control in normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density so that absorbance values can be adjusted for plating differences. The phospho-specific sub-type and Cell Death research area focus make the kit suitable for measuring relative levels of phosphorylated FADD in cultured cells and for screening the effects of treatments, inhibitors, or activators on FADD phosphorylation.

FADD (Fas-associated death domain protein) is an adaptor molecule that mediates apoptosis, recruited through its C-terminal death domain by Fas-receptor and related TNF-family receptors to engage caspase-8 and the cysteine protease cascade. Phosphorylation of FADD at Ser191 has been implicated in cellular processes including immune response and cancer development, supporting use of this kit in research on apoptosis, cell signaling, and cancer biology.
