Description

A competitive ELISA kit from Assay Genie for the quantitative measurement of folic acid (Vitamin B9) in biological samples, supporting research into this essential nutrient's roles in DNA synthesis, cell growth, and metabolism. Folic acid deficiency has been linked to health concerns including anemia, birth defects, and cardiovascular diseases, and the kit is intended for use across research into folic acid's involvement in physiological and pathological conditions.

The assay uses a colorimetric detection method in a competitive ELISA format coated with antigen. Reported sensitivity is 46.875 pg/mL with a working range of 78.125–5000 pg/mL. The kit is reactive across universal sample types and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Storage is at 2–8°C (sealed) and the components should not be cryopreserved.

The procedure adds 50 µL of standard or sample together with 50 µL of biotin-labelled antibody per well, with incubation at 37°C for 45 minutes, followed by washing, addition of TMB substrate with incubation in the dark at 37°C for 10–20 minutes, addition of stop solution (with colour change from blue to yellow), and absorbance measurement at 450 nm on a microplate reader. Reported linearity in serum, EDTA plasma and heparin plasma across 1:2, 1:4 and 1:8 dilutions ranges from 82% to 104%, and recovery in the same matrices ranges from 85% to 105% with averages of 92–96%. Sample preparation guidance includes clotting and centrifugation of serum at 1000 × g for 20 minutes, and collection of EDTA plasma by centrifugation at 1000 × g for 15 minutes at 2–8°C, with similar centrifugation-based protocols for cell culture supernatant, cell lysate and tissue homogenate.
