Estrogen Receptor-beta (ERβ, ESR2) is a nuclear hormone receptor that acts as a ligand-activated transcription factor, binding estrogen response elements (EREs) in promoter DNA to drive gene expression in reproductive, neural, cardiovascular, and immune tissues. This DNA-binding ELISA from Assay Genie quantitatively measures ERβ transcriptional activity in nuclear extracts prepared from human, mouse, or rat cells. The microplate is pre-coated with a consensus ERE oligonucleotide that captures activated receptor complexes; bound ERβ is then detected with a receptor-specific antibody in an absorbance-based readout. The protocol uses a 20 µL sample of nuclear lysate per well and runs in approximately 3.5 hours, providing a practical tool for comparing ERβ activation across hormone treatments, knock-down experiments, and screening of endocrine-active compounds. Reagents should be stored at 4 °C, with the antibody vials aliquoted and held at -20 °C for longer term to limit freeze-thaw damage; the kit is intended for research use only and supports studies of estrogen signaling, hormone-dependent cancer biology, and nuclear-receptor pharmacology.
