Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the accurate detection of Estrogen Receptor-beta levels in cell culture samples. The colorimetric assay uses a 96-Well Microplate format and is validated for reactivity with Human, Mouse and Rat samples. It is suited to adherent cells and offers a dynamic range of > 5000 Cells, with absorbance read at 450 nm.

The assay operates as a high-throughput, lysate-free indirect ELISA. Estrogen Receptor-beta is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, enabling a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density, allowing absorbance values to be normalised to cell amounts.

The Estrogen Receptor-beta target (gene symbol ESR2, Gene ID 2100, UniProt ID Q92731) is a nuclear hormone receptor and transcription factor that binds estrogens with an affinity similar to ER-alpha and regulates gene expression in target tissues. Dysregulation of Estrogen Receptor-beta signalling has been linked to diseases such as breast cancer, osteoporosis and cardiovascular disorders, and the kit is intended for research applications including measuring relative Estrogen Receptor-beta levels in cultured cells and screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators. Storage is at 4°C with a stability of 6 Months.
