Description

From Assay Genie, this cell-based ELISA kit is designed to detect and quantify phosphorylation of estrogen receptor alpha at serine 104 in cultured cells. The qualitative, indirect ELISA format uses phospho-specific primary antibodies to capture the target and HRP-conjugated secondary antibodies for detection, with colorimetric readout at 450 nm. The assay is described as convenient, lysate-free, high-throughput and sensitive, allowing measurement of relative levels of phosphorylated estrogen receptor alpha in cultured cells and screening of the effects that treatments, inhibitors (such as siRNA or chemicals) or activators have on receptor phosphorylation.

Reactivity is reported for human, mouse and rat samples, with a dynamic range of > 5000 cells. The intended sample type is adherent cells. The target is the ESR1 gene product (Gene ID: 2099; UniProt ID: P03372), a nuclear hormone receptor that regulates eukaryotic gene expression and affects cellular proliferation and differentiation. Phosphorylation at serine 104 has been shown to impact transcriptional activity and downstream signaling, making the modification an important biomarker for studies of hormone-related diseases such as breast cancer and osteoporosis.

For data normalization, the kit includes a monoclonal antibody specific for human GAPDH as an internal positive control to normalize target absorbance values, and supports a Crystal Violet whole-cell staining method after HRP-substrate measurement to determine cell density and correct for plating differences.
