Estrogen Receptor-alpha Colorimetric Cell-Based ELISA Kit

Designed by Assay Genie for accurate measurement of estrogen receptor alpha in cell culture samples, this lysate-free, cell-based colorimetric ELISA offers a high-throughput workflow. The kit detects Estrogen Receptor-alpha protein expression profiles in adherent cells, with a dynamic range of > 5000 Cells. It can be used to measure the relative amounts of Estrogen Receptor-alpha in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on the target. Reactivity has been validated for Human and Mouse samples.

Qualitative determination of Estrogen Receptor-alpha concentration is achieved via an indirect ELISA format: target protein is captured by Estrogen Receptor-alpha-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The conjugated HRP enzyme catalyses a colorimetric reaction measured at 450 nm upon substrate addition. Because the assay is qualitative, two normalization methods are included: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method can be used to determine cell density, enabling absorbance values to be normalized to cell amounts and correcting for plating differences.
