Estradiol ELISA Kit

This competitive ELISA kit from Assay Genie enables quantitative measurement of Estradiol in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a double-antigen sandwich format on a pre-coated microplate, with biotin-labeled antigen forming a sandwich complex, HRP-conjugated Streptavidin and TMB substrate producing a colorimetric reaction read at 450 nm. Sensitivity is 7.5 pg/mL with a measuring range of 12.5–800 pg/mL, reactivity is described as universal, and the kit should be stored sealed at 2–8°C without cryopreservation. Linearity was validated at 1:2, 1:4 and 1:8 dilutions: serum (n=10) 86–100%, 82–96%, 83–98%; EDTA plasma (n=10) 85–105%, 83–98%, 86–99%; heparin plasma (n=10) 84–100%, 81–97%, 93–104%. Recovery ranges with averages are serum (n=10) 87–105% (95%), EDTA plasma (n=10) 85–104% (92%), and heparin plasma (n=10) 88–103% (95%). The protocol calls for equilibrating TMB substrate for 30 minutes at room temperature, adding 50 µL of standard or sample with 50 µL of biotin-labeled antibody per well, incubating at 37°C for 45 minutes, washing five times with 1-minute soaks, developing in the dark at 37°C for 10–20 minutes, stopping the reaction, and reading absorbance at 450 nm.
