The ERF Transcription Factor Activity Assay from Assay Genie is a DNA-binding ELISA built to measure the DNA-binding activity of ETS2 repressor factor (ERF), a member of the ETS transcription factor family that functions as a transcriptional repressor on ETS-responsive promoters. The plate is pre-coated with a double-stranded oligonucleotide containing the ERF/ETS core consensus motif (5'-GGAA-3'), and active ERF in nuclear or cell lysates is captured during incubation. Bound complexes are detected with an ERF-specific primary antibody, an HRP-conjugated secondary antibody, and a TMB substrate, yielding a quantitative readout of binding activity rather than total protein abundance.

Each kit provides reagents sufficient for 96 Assays, validated specifically for human, mouse, and rat nuclear extracts. Researchers investigating MAPK/ERK signaling, tumor suppression, developmental gene regulation, or ETS-family transcriptional networks can use this assay to compare ERF binding activity across treated versus untreated conditions, time-course experiments, or different cell lines. The assay provides a functional complement to standard western blot analyses of ERF expression by directly reporting DNA-binding competence rather than total protein levels.

For reliable results, refrigerate all kit components at 4 °C, avoid repeated freeze-thaw cycles of the antibody and oligonucleotide reagents, and store nuclear extracts in single-use aliquots. Allow samples to equilibrate before running the assay. This kit is intended for research use only and is not validated for diagnostic or clinical applications.
