Description

A cell-based ELISA kit from Assay Genie designed for the qualitative measurement of ERCC5 protein expression in cultured human cells, using an adherent cell sample type. The kit uses an indirect ELISA format in which ERCC5 is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies, with a colorimetric readout at 450 nm. It is described as lysate-free and high-throughput, and is suitable for measuring relative ERCC5 amounts in cultured cells as well as for screening the effects of treatments, inhibitors such as siRNA or chemicals, or activators on ERCC5 levels.

The dynamic range is greater than 5000 cells per well. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density so that absorbance values can be normalized to cell amounts. Storage stability is 4 °C for 6 months.

ERCC5, also known as Xeroderma Pigmentosum group G (XPG) protein, is a single-strand-specific DNA endonuclease that makes the 3' incision in DNA nucleotide excision repair and plays a role in DNA repair mechanisms and the maintenance of genomic stability. Dysregulation of ERCC5 has been linked to a range of genetic disorders and diseases, including cancer and neurological disorders.
