Description
Assay Genie provides a cell-based ELISA kit for measuring phosphorylation of the Erythropoietin Receptor (Epo-R) at tyrosine 368 in adherent cells. The colorimetric assay is read at 450 nm and shows reactivity with human, mouse, and rat samples, with a stated dynamic range of > 5000 cells. Detection uses an indirect ELISA format in which phospho-Epo-R is captured by target-specific primary antibodies, with HRP-conjugated secondary antibodies catalysing the colorimetric reaction upon substrate addition.
A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density and correct for plating differences. The kit is intended for measuring relative levels of phosphorylated Epo-R in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on Epo-R phosphorylation. According to Assay Genie, it is suitable for basic research, drug discovery, and clinical diagnostics, and is relevant to studies of erythropoietin signalling, hematopoietic disorders, and certain cancers. The target protein is encoded by the EPOR gene (Gene ID: 2057, UniProt ID: P19235).
