Description

This colorimetric cell-based ELISA kit from Assay Genie is designed for the accurate quantification of phosphorylated EPB41 protein levels in cell lysates and tissue samples. The kit specifically targets EPB41 phosphorylation at tyrosine residues Tyr660/418 and is reactive with human and mouse samples. Detection is performed at 450 nm in an indirect ELISA format in which phospho-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze the colorimetric reaction.

Described as lysate-free, high-throughput, and sensitive, the assay can be used both to measure relative levels of phosphorylated EPB41 in cultured cells and to screen the effects that various treatments, inhibitors, or activators have on EPB41 phosphorylation. A monoclonal GAPDH antibody is included as an internal positive control for normalizing target absorbance, and the Crystal Violet whole-cell staining method can be used to determine cell density and adjust for plating differences. The kit has a dynamic range of more than 5000 cells.

EPB41, also known as protein 4.1 or erythrocyte membrane protein band 4.1, is a major structural element of the erythrocyte membrane skeleton that stabilizes the spectrin–actin interaction and contributes to red cell mechanical stability and deformability. It also recruits DLG1 to membranes. Phosphorylation at Tyr660/418 regulates EPB41 activity, and dysregulation of this modification has been linked to cancer, neurological disorders, and autoimmune conditions. The protein has a reported molecular weight of 97 kDa, with Gene ID 2035 and UniProt ID P11171; mutations in the gene are associated with type 1 elliptocytosis (EL1).
