Description

A cell-based ELISA from Assay Genie is designed for the sensitive and specific detection of phosphorylated ELK1 at serine 383 in cell lysates and tissue homogenates, supporting research into cell signaling and transcription regulation. Phosphorylation of ELK1 at this residue is known to modulate its transcriptional activity, making it relevant to studies of cancer, inflammation, and neurological disorders.

Detection is performed by an indirect ELISA in which phospho-specific primary antibodies capture ELK1 (Phospho-Ser383) and HRP-conjugated secondary antibodies catalyze a colorimetric reaction measured at 450 nm. The assay is lysate-free and suitable for high-throughput screening of treatments, inhibitors, or activators on ELK1 phosphorylation. It is reactive with human, mouse, and rat samples, with a stated dynamic range of > 5000 Cells. An included anti-GAPDH monoclonal antibody serves as an internal positive control for normalization, and a Crystal Violet whole-cell staining step allows determination of cell density for further normalization.

ELK1 is a transcription factor of the Ets family and ternary complex factor (TCF) subfamily. It forms a ternary complex by binding the serum response factor at the serum response element in the promoter of the c-fos proto-oncogene and is phosphorylated by MAP kinase pathways at a cluster of S/T motifs at its C-terminus, an event critical for transcriptional activation. Associated database entries include Gene ID 2002 and UniProt ID P19419. The kit is categorized under the epigenetics and nuclear signaling research area.
