Description

This cell-based ELISA kit from Assay Genie is designed for the qualitative determination of phosphorylated eukaryotic translation initiation factor 4B (eIF4B) at Serine 422 in cell lysates. The assay uses a colorimetric method with readout at 450 nm in an indirect ELISA format employing HRP-conjugated secondary antibodies, and is reactive with human and mouse samples. It is described as a convenient, lysate-free, high-throughput and sensitive approach for measuring relative levels of phosphorylated eIF4B in cultured adherent cells and for screening the effects of treatments, inhibitors or activators on eIF4B phosphorylation.

The kit is provided in a 96-well microplate format and falls within the Metabolism research area, with a phospho-specific target. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining step is used to determine cell density so that absorbance values can be adjusted to cell amounts. The stated dynamic range is > 5000 cells.

EIF4B is a key regulator of protein synthesis, required for the binding of mRNA to ribosomes and functioning in close association with eIF4-F and eIF4-A. Phosphorylation at Serine 422 is reported to play a critical role in the control of protein synthesis and is described as dysregulated in diseases such as cancer and metabolic disorders, supporting its use as a biomarker for studying disease mechanisms and targeted therapies.
