Description

A cell-based ELISA kit from Assay Genie designed for the qualitative measurement of EGFR (Epidermal Growth Factor Receptor) phosphorylation at tyrosine 1092 in cultured cells. The assay uses an indirect ELISA format: phospho-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to produce a colourimetric reaction upon substrate addition, yielding colourimetric detection at 450 nm. It is suitable for measuring relative phosphorylation levels and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on EGFR phosphorylation. Sample type is adherent cells and reactivity covers Human, Mouse and Rat.

Two approaches to data normalisation are provided. A monoclonal antibody specific for human GAPDH serves as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining step may be used to determine cell density, allowing absorbance values to be normalised to cell number. The stated dynamic range is greater than 5000 cells.

EGFR is a receptor tyrosine kinase that regulates cell growth, survival and differentiation, with phosphorylation at tyrosine 1092 implicated in the activation of downstream signalling pathways. Dysregulation of EGFR signalling is associated with cancer development and progression, making the kit a useful tool for studying EGFR signalling pathways and potential therapeutic targets in cancer research.
