Description

Assay Genie supplies this Colorimetric Cell-Based ELISA Kit for measuring phosphorylated EGFR at Tyr1016 in cell lysates. The assay uses adherent cells in an indirect ELISA format, in which a phospho-Tyr1016-specific primary antibody is bound and then detected with an HRP-conjugated secondary antibody, producing a colorimetric readout at 450 nm. The kit is reactive with Human, Mouse and Rat samples and has a stated dynamic range of > 5000 Cells. It is described as a lysate-free, high-throughput and sensitive assay suitable for measuring relative EGFR phosphorylation in cultured cells and for screening the effects of various treatments, inhibitors (e.g. siRNA or chemicals) or activators on EGFR phosphorylation.

A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values. Following colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, and absorbance values can then be normalized to cell amounts to adjust for plating differences.

EGFR (Epidermal Growth Factor Receptor) is a receptor tyrosine kinase involved in the control of cell growth and differentiation; phosphorylation of specific tyrosine residues such as Tyr1016 is associated with receptor activation and downstream signaling, and dysregulation of EGFR signaling has been implicated in diseases including cancer. Related database identifiers listed for the target include Gene ID 1956 and UniProt ID P00533.
