Description

The Assay Genie Phospho-EGFR (Thr693) Fluorometric Cell-Based ELISA Kit is a lysate-free, high-throughput assay designed to monitor EGFR phosphorylation and the expression profile of EGFR in cells. Using an indirect ELISA format, the kit enables qualitative measurement of relative amounts of phosphorylated EGFR in cultured cells and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on EGFR phosphorylation.

Target-specific primary antibodies are paired with Dye 1- and Dye 2-conjugated secondary antibodies, allowing fluorometric detection read at Ex/Em: 651/667 (Dye 1) and 495/521 (Dye 2). A monoclonal antibody specific for human GAPDH is included as an internal positive control, and an antibody against the non-phosphorylated counterpart of EGFR (Phospho-Thr693) is provided so that RFU values for non-phosphorylated EGFR can be used to normalize the RFU value for phosphorylated EGFR. The kit is supplied in a 96-well black cell culture clear-bottom microplate format, is reactive with Human, Mouse, and Rat samples, has a dynamic range of > 5000 Cells, and is stored at 4°C for 6 months.

EGFR is a receptor tyrosine kinase that binds epidermal growth factor and related ligands, with ligand binding triggering receptor dimerization, autophosphorylation, and downstream signalling. Phosphorylation of EGFR on Ser and Thr residues is thought to represent a mechanism for attenuation of EGFR kinase activity. The target is also referenced in databases under Gene ID 1956 and UniProt ID P00533.
