EFNA5 Colorimetric Cell-Based ELISA

A cell-based ELISA kit from Assay Genie designed for the qualitative detection of EFNA5 (Ephrin-A5) protein expression directly in cultured cells, without the need for lysate preparation. The assay uses a colorimetric detection method measured at 450 nm and has been validated for human, mouse, and rat reactivity. It is supplied in a 96-well microplate format, is intended for use with adherent cell samples, and supports a dynamic range of > 5000 cells.

The workflow follows an indirect ELISA format, in which EFNA5 is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for data normalization, and a Crystal Violet whole-cell staining step can be used to determine cell density and correct for plating variation. Beyond straightforward quantification, the kit is suited to screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on EFNA5 expression in cells.

EFNA5 is a GPI-anchored cell surface ligand for Eph receptors, with roles in cell signaling relevant to neuronal, vascular, and epithelial development, including axon guidance and the regulation of cell-cell adhesion. The kit is stored at 4 °C with a stated stability of 6 months.
