Description

Designed for the accurate detection of Eukaryotic Elongation Factor 2 Kinase (EEF2K), this colorimetric cell-based ELISA kit from Assay Genie provides a lysate-free, high-throughput workflow for measuring relative target protein levels directly in cultured adherent cells. The assay supports qualitative determination of EEF2K concentration and screening for the effects of treatments, inhibitors (such as siRNA or chemicals) or activators, using an indirect ELISA format in which EEF2K is captured by target-specific primary antibodies and detected by HRP-conjugated secondary antibodies that catalyse a colorimetric reaction upon substrate addition.

The kit is reactive with human and rat samples, with absorbance read at 450 nm, a stated dynamic range of > 5000 cells, and storage stability of 4 °C for 6 months. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining method is supplied for determining cell density so that absorbance readings can be normalised to cell amount and plating differences can be adjusted.

EEF2K is an atypical calcium/calmodulin-dependent protein kinase that phosphorylates and inactivates eukaryotic elongation factor 2, linking cellular energy status to the regulation of protein synthesis and influencing cell proliferation, apoptosis and autophagy. Dysregulation has been linked to cancer, neurodegenerative disorders and metabolic disease, and the target is catalogued under NCBI Gene ID 29904, UniProt ID O00418 and OMIM 606968.
