Description

Designed by Assay Genie for the precise measurement of EEF2 phosphorylation at threonine 56 in cell lysates, this Colorimetric Cell-Based ELISA Kit is intended for use with adherent cells and enables researchers to study protein synthesis regulation and related cell signalling pathways. The kit targets eEF2 (Phospho-Thr56) and is reactive with human, mouse, and rat samples, supporting a broad range of research applications with a dynamic range of > 5000 Cells.

The assay uses a lysate-free, high-throughput indirect ELISA format in which eEF2 (Phospho-Thr56) is captured by phospho-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyse a colorimetric reaction read at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalise target absorbance values, and Crystal Violet whole-cell staining is provided for determining cell density and adjusting absorbance readings accordingly. The kit can be used to measure relative levels of phosphorylated eEF2 in cultured cells and to screen for the effects of various treatments, inhibitors such as siRNA or chemicals, or activators on eEF2 phosphorylation.

EEF2, or eukaryotic elongation factor 2, is an essential GTP-binding factor that promotes GTP-dependent translocation of the nascent protein chain from the A-site to the P-site of the ribosome during translation. Phosphorylation at threonine 56 is a key regulatory event that can impact translation efficiency and cellular processes, with altered EEF2 phosphorylation linked to conditions including cancer and neurodegenerative disorders, making this kit a useful resource for studying disease mechanisms.
