The CD66b positive selection kit from STEMCELL Technologies isolates granulocytes directly from whole blood for HLA chimerism studies, supporting lineage-specific engraftment monitoring in hematopoietic stem cell transplantation research. CD66b is a GPI-anchored surface glycoprotein expressed on granulocytes, allowing researchers to resolve myeloid-lineage chimerism independently of lymphoid-lineage results when whole-blood values are ambiguous or mixed.

Built on the EasySep immunomagnetic platform, the kit uses antibody complexes that label target cells in suspension without density-gradient centrifugation or columns. Anticoagulated whole blood is processed in a 15 mL tube: the antibody cocktail is added, briefly incubated, and the tube is placed in an EasySep magnet so unlabeled cells can be decanted while the labeled CD66b-positive fraction is retained. The resulting granulocyte preparation typically reaches >95% purity and is supplied in a single-use, research-use-only kit format suitable for downstream DNA extraction, PCR amplification of HLA loci, and chimerism quantification by STR or qPCR.

Applications include tracking donor myeloid reconstitution after allogeneic HSCT, distinguishing graft rejection from relapse in mixed chimerism patients, and supporting graft-versus-host disease studies where neutrophil recovery kinetics are clinically relevant. Components are refrigerated on receipt to preserve reagent activity, and the workflow integrates with existing chimerism analysis pipelines used by transplant immunology laboratories.
