EasySep Human Mono Iso Kit

From STEMCELL Technologies, this EasySep™ kit enables efficient isolation of highly purified human monocytes from PBMCs using a simple, optimized immunomagnetic protocol that can be completed in as little as 12 minutes. Starting with PBMCs from human peripheral blood, the typical monocyte cell content (CD14+CD16-) of the isolated fraction is 89.7 ± 3.4% using the purple EasySep™ Magnet. When combined with the EasySep™ Human Platelet Removal Cocktail, the typical purity is 87.3 ± 4.5%.

Isolated monocytes retain the capacity to differentiate into mature dendritic cells and macrophages in downstream culture. In validation studies, mature dendritic cells generated from kit-isolated monocytes expressed the CD83 marker on 93 ± 5% of cells at Day 7 (with 1 ± 1% still expressing CD14), yielding 45 ± 25% viable mature DCs, and secreted IL-12 (p70) at 361 ± 81 pg/mL upon activation. The kit also preserves monocyte-to-macrophage and monocyte-to-dendritic-cell differentiation efficiency when isolated from refrigerated leukopak fractions stored for up to 5 days.
