EasySep Rat IgM Depletion Kit

This immunomagnetic positive selection kit from STEMCELL Technologies enables rapid, column-free depletion of IgM+ cells from single-cell suspensions of rat splenocytes, lymph nodes, or whole blood. The procedure is fast and easy to use, and because it targets unwanted cells for removal, the isolated cells are left untouched and immediately ready for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction. Up to 90% purity (≤ 10% IgM+) can be achieved.

The kit comprises the EasySep™ Rat IgM Depletion Cocktail (a combination of monoclonal antibodies in PBS and 0.1% BSA), EasySep™ Dextran RapidSpheres™ (a suspension of magnetic particles in water), and an optional Anti-Rat CD32 Blocker (monoclonal antibodies in PBS, 0.1% BSA, and < 0.1% sodium azide). All components should be stored at 2 - 8°C, must not be frozen, and remain stable until the expiry date on the label; they may be shipped at room temperature (15 - 25°C). Recommended media include EasySep™ Buffer, RoboSep™ Buffer, or PBS containing 2% FBS and 1 mM EDTA, free of Ca++ and Mg++.

The kit has been validated with Sprague Dawley and Wistar rat strains and is expected to be compatible with other strains. In a representative experiment using a single-cell suspension of splenocytes from a Sprague Dawley rat, the IgM content of the final isolated fraction was typically 8.0 ± 4.1%, with start and final depleted fractions of 55.7% and 8.7% respectively, representing a 1.6 log depletion.
