Description

An In-Cell ELISA Kit manufactured by Assay Genie, designed for the qualitative detection of DYRK1A (dual-specificity tyrosine-phosphorylation-regulated kinase 1A) protein levels in cultured cells. The assay uses a colorimetric detection method at 450 nm and is suitable for use with adherent cells, with demonstrated reactivity against Human, Mouse, and Rat samples. It provides a lysate-free, high-throughput workflow in a 96-well microplate format, with a stated dynamic range of > 5000 cells.

Detection is achieved via an indirect ELISA format: the target is bound by DYR1A-specific primary antibodies, which are then detected by HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density and adjust for plating differences. The kit can be applied to measuring relative DYR1A expression in cultured cells and to screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on DYR1A levels.

DYRK1A is a dual-specificity protein kinase of the DYRK family, localised in the Down syndrome critical region of chromosome 21 (21q22.13), with documented roles in cell proliferation, brain development, and circadian rhythm, and associations with developmental disorders, neurodegenerative disease, and cancer. The kit is stored at 4 °C with a stability of 6 months and is intended for research applications in cell biology, neuroscience, and oncology.
