Description

This phospho-specific cell-based ELISA kit from Assay Genie is designed for the detection of Dynamin-1 phosphorylated at Ser774. It targets the dynamin-1 protein, encoded by the DNM1 gene, with reactivity against human, mouse, and rat samples. The assay is intended for use with adherent cells and provides a dynamic range of greater than 5000 cells using a colorimetric detection method at 450 nm.

The kit uses an indirect ELISA format in which Dynamin-1 (Phospho-Ser774) is captured by phospho-specific primary antibodies. HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, while Crystal Violet whole-cell staining can be used to determine cell density and adjust for plating differences. Researchers can use the assay to measure relative amounts of phosphorylated Dynamin-1 in cultured cells or to screen for the effects of various treatments, inhibitors such as siRNA or chemicals, and activators on Dynamin-1 phosphorylation.

Dynamin-1 is a microtubule-associated GTP-binding protein involved in vesicular trafficking, clathrin-mediated endocytosis, and synaptic vesicle recycling. Phosphorylation of Dynamin-1 at Ser774 modulates its function and is implicated in neurological disorders such as Alzheimer's disease and Parkinson's disease, making this kit relevant for cell biology research into Dynamin-1 regulatory mechanisms and disease pathology.
