DUSP4 Colorimetric Cell-Based ELISA

A cell-based ELISA kit from Assay Genie is designed for the detection of Dual Specificity Phosphatase 4 (DUSP4) in adherent cells, with reactivity against human, mouse, and rat samples. The assay uses a colorimetric detection method measured at 450 nm and is provided in a 96-well microplate format. The kit is lysate-free, high-throughput, and sensitive, allowing researchers to study the regulatory effects of DUSP4 on cellular signaling pathways, including its role as a critical regulator of MAP kinase signaling that inactivates ERK1, ERK2, and JNK. DUSP4 controls the activity of transcription factors involved in cellular response to external stimuli, and dysregulation of DUSP4 has been linked to diseases including cancer and inflammatory disorders.

The assay uses an indirect ELISA format in which DUSP4 is captured by DUSP4-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, enabling a colorimetric reaction upon substrate addition. The kit can be used to measure relative amounts of DUSP4 in cultured cells and to screen for the effects of various treatments, inhibitors such as siRNA or chemicals, or activators on DUSP4. Because the assay is qualitative, two normalization methods are included: a monoclonal anti-GAPDH antibody serves as an internal positive control, and a Crystal Violet whole-cell staining step is used to determine cell density and normalize absorbance values to plating differences.

For storage, the kit is stable at 4°C for 6 months.
