Description

Designed for the quantitative detection of DOK7 protein levels in cell lysates, this Cell-Based ELISA kit from Assay Genie uses a colorimetric detection method read at 450 nm. The assay is reactive with human and mouse samples and is suited to adherent cells. It functions as a lysate-free, high-throughput, and sensitive format for measuring relative DOK7 expression in cultured cells, as well as for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on DOK7.

Qualitative determination of DOK7 concentration is achieved via an indirect ELISA format: DOK7 is captured by DOK7-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. Because the assay is qualitative, two normalisation approaches are included: a monoclonal antibody specific for human GAPDH serves as an internal positive control, and a Crystal Violet whole-cell staining method may be used to determine cell density so that absorbance values can be normalised to cell amounts. The dynamic range is reported as > 5000 Cells, and the kit is supplied in a 96-Well Microplate format for storage at 4°C with a 6-month shelf life.

DOK7 is described as a key regulator of neuromuscular synapse development and maintenance, with a critical role in skeletal muscle function, and its dysregulation has been linked to various neuromuscular disorders, making it a valuable target for therapeutic and drug-development research. UniProt ID: Q18PE1; Gene ID: 285489.
