This Sandwich ELISA Kit from Assay Genie quantitatively measures canine PTEN in serum and plasma samples using a colorimetric detection method typically read at 450 nm. PTEN (Phosphatase and Tensin homolog) is a critical tumor suppressor that negatively regulates the PI3K/AKT signaling pathway, making it a key biomarker in veterinary oncology research, signal transduction studies, and investigations into cancer progression in canine disease models.

The assay employs a 96-well microplate pre-coated with a capture antibody, with results calculated against a standard curve. Samples and standards are incubated, washed, and treated with detection antibody and substrate to produce absorbance changes proportional to PTEN concentration. Recommended storage is at 2-8 °C, and the kit is intended for research use only.

Reagents are supplied ready-to-use or as concentrated stock depending on component, and the protocol is optimized for sensitivity and specificity across a broad dynamic range. Researchers studying mammary tumors, melanoma, lymphoma, and other neoplasms in dogs will find this microplate assay useful for biomarker quantification and pathway analysis in veterinary translational research.

PTEN functions as a lipid phosphatase that dephosphorylates PIP3 back to PIP2, directly antagonizing PI3K signaling and constraining downstream AKT and mTOR activity. Loss-of-function alterations, including mutation, copy-number deletion, and promoter methylation, have been documented in canine hemangiosarcoma, osteosarcoma, mast cell tumors, oral melanoma, and mammary carcinoma, positioning PTEN as a prognostic biomarker and therapeutic entry point in comparative oncology research supported by the Assay Genie product line.

The sandwich ELISA format uses a pre-coated 96-well strip plate paired with a biotinylated detection antibody and HRP-streptavidin conjugate. Standards are prepared from recombinant canine PTEN, and the working detection range is typically 0.312–20 ng/mL, with analytical sensitivity below 0.119 ng/mL and intra-assay CV under 10%. Up to 40 unknown canine serum or plasma samples can be quantified in duplicate alongside the standard curve, with absorbance read at 450 nm within 15 minutes of stop solution addition.

Unopened kits are stored at 2–8 °C; reconstituted standards and concentrated detection reagents should be aliquoted into vials and frozen at -20 °C to limit freeze-thaw damage. Whole blood is collected without anticoagulant interference, clotted for 30 minutes, centrifuged at 1000 x g for 15 minutes, and the resulting serum aliquoted and stored at -80 °C if not assayed within 24 hours. Heparin or EDTA plasma is acceptable, with reported spike recoveries of 87–105% across matrices, confirming compatibility with this research use only ELISA workflow for veterinary biomarker discovery and pathway analysis.
