Description
STEMCELL Technologies supplies this Animal Component-Free (ACF) DNase I preparation, an endonuclease obtained from cultures free of animal-derived materials and purified by chromatography. The enzyme is supplied as a lyophilized powder derived from Pichia pastoris and is intended for digestion of DNA in research workflows, including tissue dissociation protocols where extracellular DNA released from damaged cells can otherwise complicate downstream processing.
The enzyme consists of a single glycosylated polypeptide chain with two disulfide bonds and preferentially cleaves phosphodiester linkages adjacent to pyrimidine nucleotides in both single- and double-stranded DNA, yielding polynucleotides with 5'-phosphate and 3'-hydroxyl groups. It has a molecular weight of 29.1 kDa (CAS Number 9003-98-9) and an optimum pH of 7.8, and the stated activity is ≥ 2000 units/mg dry weight. Alternative names listed by the manufacturer include DNA endonuclease, DNA nuclease, deoxyribonucleic phosphatase, pancreatic DNase, and thymonuclease. The product may contain protease and RNase, and the final dialysis buffer prior to lyophilization is 10 mM calcium acetate, pH 5.0, containing glycine.
For use, the lyophilized material can be reconstituted in sterile water or a suitable buffer within pH 4.5–9.0, such as Hanks' Balanced Salt Solution or other non-phosphate buffers, with a recommended stock concentration of 1–10 mg/mL. Reconstituted solutions intended for cell culture should be sterilized through a 0.22 μm filter, aliquoted, and stored frozen at -20°C or -80°C; repeated freeze-thaw cycles of aliquots should be avoided. The unreconstituted product is stored at 2–8°C and is stated to be stable as supplied for 12 months from the date of receipt. According to the supplier's safety documentation, the product is not classified as a hazardous substance or mixture under the Globally Harmonized System (GHS), and is intended for research use only.
