A crude collagenase enzyme blend from STEMCELL Technologies, this Animal Component-Free (ACF) formulation is derived from Clostridium histolyticum and is intended for dissociating mammalian tissue to release viable cells for primary cell culture, flow cytometry, and single-cell sequencing workflows. The lyophilized powder, packaged in a 1 g vial, combines collagenase activity with clostripain and neutral protease activities so that extracellular matrix proteins such as type I and type III collagen are digested alongside the supporting stromal components. Researchers commonly reconstitute the powder at 10 to 100 mg/mL in a balanced salt solution or a dedicated digestion buffer and pair it with a supplemental neutral protease to tune dissociation kinetics for delicate tissues such as pancreatic islets, hepatocytes, lung parenchyma, and the stromal vascular fraction of adipose tissue. Each lot is tested for collagen digestion activity and is shipped refrigerated; reconstituted stock should be stored at -20 °C in single-use aliquots to limit freeze-thaw loss of enzymatic activity, and shelf life depends on storage temperature and handling. The Animal Component-Free designation supports xeno-free and clinically oriented workflows where animal-source contaminants must be documented as absent from raw materials used to generate cell products.
