For cell therapy and regenerative medicine workflows requiring animal-free reagents, the STEMCELL Technologies DNase I ACF formulation delivers 100,000 units of endonuclease activity in a single vial. The enzyme hydrolyzes both single- and double-stranded DNA into oligonucleotides bearing 5'-phosphate termini, and depends on divalent cations such as Ca2+ and Mg2+ for full activity. One unit is defined as the amount of enzyme that completely degrades 1 µg of plasmid DNA in 10 minutes at 37°C in standard reaction buffer. Common applications include removing extracellular DNA from cell suspensions to prevent cell clumping during tissue dissociation, clearing DNA contamination from RNA preparations used in molecular biology assays, and processing single-cell suspensions for downstream flow cytometry or sequencing workflows. Because the formulation is free of animal-derived components, it integrates into GMP-compatible manufacturing pipelines used in pluripotent stem cell maintenance, hematopoietic cell expansion, and immunotherapy research. The product is supplied in a single vial and should be stored at -20°C; aliquoting upon first use is recommended to limit freeze-thaw cycles and preserve long-term activity.
