Description

Supplied by STEMCELL Technologies as a lyophilized powder, this endonuclease is derived from bovine pancreas and is intended for the digestion of DNA. It is commonly incorporated into tissue dissociation workflows to degrade extracellular DNA released by damaged cells. The enzyme preferentially cleaves phosphodiester linkages adjacent to pyrimidine nucleotides in both single- and double-stranded DNA, yielding polynucleotides with 5'-phosphate and 3'-hydroxyl termini. It consists of a single glycosylated polypeptide chain with two disulfide bonds and has a molecular weight of 29.1 kDa (CAS Number 9003-98-9).

The reagent can be reconstituted in a balanced salt solution or buffer of choice and should be stored at 2 - 8°C, where it remains stable for 12 months from the date of receipt. Activity is specified at ≥ 2000 units/mg dry weight, with one unit defined as the amount required to act on 1 mg/mL of DNA at pH 5.0 and 25°C to produce an increase in absorbance of 0.001 per minute at a wavelength of 260 nm. Published reports describe its use in dissociating human microglia, cartilage, colon, epithelium, liver, lung, and neural tissues, as well as stem cells from sources such as human term placenta and the decidua.
