Description

This cell-based ELISA kit from Assay Genie is designed for the qualitative detection of DNA polymerase alpha protein expression in cultured cells. It uses an indirect ELISA format in which DNA polymerase alpha-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. The lysate-free kit is intended for high-throughput screening of relative DNA polymerase alpha levels in cells and for evaluating the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on the target.

The assay is reactive with human samples and is performed on adherent cells. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values. Following the colorimetric HRP readout, a Crystal Violet whole-cell staining step may be used to determine cell density and adjust for plating differences. Detection is by colorimetric measurement at 450 nm using a 96-well microplate format, with a dynamic range of > 5000 cells. The product is stored at 4 °C with a stated stability of 6 months.

DNA polymerase alpha plays an essential role in the initiation of DNA replication and is also involved in DNA repair processes that help maintain genomic stability. Dysregulation of its activity has been linked to diseases including cancer and genetic disorders, making it a relevant target for research into disease mechanisms and therapeutic development. The target is encoded by the POLA1 gene (Gene ID: 5422; UniProt ID: P09884).
