Description

This animal component-free neutral protease (also known as a non-specific endoprotease or proteinase) from STEMCELL Technologies is supplied as a lyophilized powder. It is obtained from Bacillus polymyxa cultures free of animal-derived materials, with a reported molecular weight of 32.5 kDa and CAS Number 42613-33-2. The enzyme cleaves the N-terminal peptide bond of non-polar amino acid residues (X/Y = nonspecific) and exhibits mild proteolytic activity over an optimum pH range of 5.9–7.0.

Activity is specified at ≥4 units/mg dry weight, where one unit releases 1 µmol folin-positive amino acids equivalent to 1 µmol tyrosine/minute from casein at 37°C, pH 7.5. Because of its mild proteolytic activity, the enzyme is particularly useful for isolating and passaging primary cells while maintaining membrane integrity, and is commonly combined with collagenase to dissociate neural, kidney, epithelial, endothelial, lung, and colon tissues, as well as stem cells. It can also serve as a secondary enzyme in cell isolation and tissue dissociation workflows. The lyophilized reagent is stable as supplied for 12 months from date of receipt and should be stored at 2–8°C; it can be reconstituted in a balanced salt solution or buffer of choice.

According to the safety data sheet, the solid powder is classified as causing skin irritation, serious eye irritation, and respiratory sensitization, and may cause allergy or asthma symptoms or breathing difficulties if inhaled. Recommended handling includes wearing protective gloves and eye/face protection, avoiding breathing dust, and ensuring adequate ventilation or respiratory protection if exposure limits are exceeded.
