A neutral metalloprotease preparation from STEMCELL Technologies, supplied at 5 U/mL activity in a 100 mL bottle, supports gentle cell dissociation workflows in research laboratories. The enzyme, isolated from Bacillus polymyxa, cleaves fibronectin and type IV collagen at the basal lamina, enabling detachment of adherent cells from primary tissue explants and confluent monolayers without damaging surface antigens or compromising viability.
Common laboratory uses include isolation of epithelial sheets from skin biopsies for keratinocyte culture, harvest of human embryonic stem cell colonies while preserving pluripotency markers, and subculture of fragile adherent lines that would otherwise be lost to trypsin digestion. A typical protocol involves a 20-40 minute incubation at 37°C followed by neutralization through dilution in standard wash medium.
For storage, keep the bottle at -20°C to preserve enzymatic activity, aliquot on receipt, and limit freeze-thaw cycles by drawing working volumes as needed for individual experiments. The single-use bottle format allows the user to dispense fresh solution for each dissociation run, supporting reproducible results in cell culture protocols.
