Description

Designed as a convenient, lysate-free, high-throughput assay from Assay Genie, this Cell-Based ELISA kit enables measurement of the relative amounts of Dematin protein expression in cultured cells. The qualitative assay uses an indirect ELISA format in which Dematin-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. Substrate addition produces a colorimetric reaction read at 450 nm, allowing screening of the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on Dematin levels in cell lysates and culture supernatants.

For data normalization, a monoclonal antibody specific for human GAPDH is included as an internal positive control. Following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, and absorbance values can then be normalized to cell amounts to adjust for plating differences. The kit has a dynamic range of > 5000 Cells and is reactive with Human and Mouse samples.

Dematin is an actin binding and bundling protein that plays a structural role in regulating the organization and stability of the cell membrane, as well as cell shape and motility. Dysregulation of Dematin expression has been implicated in various diseases, including cancer, neurological disorders, and autoimmune conditions, making it a valuable biomarker for studying these diseases and potential therapeutic interventions.
