Description

From Assay Genie, this Cell-Based ELISA Kit is a lysate-free, high-throughput tool for detecting phosphorylation of DAXX at serine 668 in adherent cell samples, with reactivity confirmed for human cells. The colorimetric detection method is read at 450 nm and the assay supports a dynamic range of > 5000 Cells.

Qualitative determination of Daxx (Phospho-Ser668) is achieved via an indirect ELISA format in which phospho-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyse a colour reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalisation, and Crystal Violet whole-cell staining can be used to determine cell density, allowing absorbance values to be adjusted for plating differences. The kit can be used to measure relative amounts of phosphorylated Daxx in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on Daxx phosphorylation.

DAXX is a transcriptional co-regulatory protein involved in apoptosis, DNA damage response and transcriptional regulation, with phosphorylation at serine 668 implicated in modulating its function and interactions with other proteins. Associated database identifiers are Gene ID 1616 and UniProt ID Q9UER7, and the assay is configured as a phospho-specific sub-type.
