Description

A Cell-Based ELISA Kit from Assay Genie designed for the detection and quantification of DARPP-32 phosphorylated at threonine 34 in cultured cells. The kit is a lysate-free, high-throughput assay using an indirect ELISA format, in which DARPP-32 (Phospho-Thr34) is captured by phospho-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. Adherent cells are the indicated sample type.

The assay is reactive with Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells and detection read at 450 nm. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalising target absorbance values, while Crystal Violet whole-cell staining can be used to determine cell density and adjust for plating differences. The workflow also supports screening of the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on DARPP-32 phosphorylation.

DARPP-32 (gene symbol PPP1R1B; UniProt Q9UD71) is a dopamine- and cyclic AMP-regulated neuronal phosphoprotein and a member of the protein phosphatase inhibitor 1 family. Phosphorylation at threonine 34 has been linked to neuronal signalling and neurotransmission and is associated with conditions such as schizophrenia, bipolar disorder, and addiction, making this phospho-specific target relevant to research in those areas.
