Description

This cell-based ELISA kit from Assay Genie is designed to measure DARPP-32 protein levels in cultured cells, cell lysates, and tissue homogenates, supporting research into dopamine neurotransmission and related neuropsychiatric disorders. The kit uses an indirect ELISA format in which DARPP-32 is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyse a colour-forming reaction.

Validated for human, mouse, and rat reactivity, the colourimetric assay is read at 450 nm and has a stated dynamic range of > 5000 Cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance, while Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating variation. The kit is supplied in a microplate format intended for storage at 4°C with a stated stability of 6 Months.

DARPP-32, also known as Dopamine- and cAMP-regulated neuronal phosphoprotein, is a member of the protein phosphatase inhibitor 1 family encoded by the PPP1R1B gene (UniProt Q9UD71). Its phosphorylation state is regulated in opposing directions by dopaminergic D1 receptor stimulation and glutamatergic NMDA receptor signalling, and dysregulation of DARPP-32 has been implicated in schizophrenia and Parkinson's disease.
